Predictive values of plasma TNFα and IL-8 for intracranial hemorrhage in patients with acute promyelocytic

Abstract: In patients with acute promyelocytic leukemia (APL), intracranial hemorrhage (ICH), if not identified promptly, could be fatal. It is the leading cause of failure of induction and early death. Thus, biomarkers that could promptly predict severe complications are critical. Here, cytokine differences between patients with APL with and without ICH were investigated to develop predictive models for this complication. The initial cytokine profiling using plasma samples from 39 patients and 18 healthy donors found a series of cytokines that were remarkedly different between patients with APL and healthy controls. The APL patients were subsequently divided into high and low white blood cell count groups. Results showed that tumor necrosis factor α and interleukin 8 (IL-8) were vital in distinguishing patients with APL who did or did not develop ICH. In addition, verification in 81 patients with APL demonstrated that the two cytokines were positively correlated with the cumulative incidence of ICH. Finally, in-vitro and in-vivo experimental evidence were provided to show that IL-8 influenced the migration of APL-derived NB4 cells and impaired the blood–brain barrier in PML/RARα positive blast-transplanted FVB/NJ mice. These assessments may facilitate the early warning of ICH and reduce future mortality levels in APL.

Abstract: Traditional on-site fault diagnosis means cannot meet the needs of large rotating machinery for its performance and complexity. Remote monitoring and diagnosis technology is a new fault diagnosis mode combining computer technology, communication technology, and fault diagnosis technology. The designed remote monitoring and diagnosis and prediction system for large rotating machinery integrates the distributed resources in different places and breaks through shortcomings as the offline and decentralized information. The system can make further implementation of equipment prediction technology research based on condition monitoring and fault diagnosis, provide on-site analysis results, and carry out online actual verification of the results. The system monitors real-time condition of the equipment and achieves early fault prediction with great significance to guarantee safe operation, saves maintenance costs, and improves utilization and management of the equipment.

Self-generation of machining precision and its realization in lapping of super precision solid balls

Abstract: This paper presents the principle of self-generation of machining precision by explaining its basic concept and five necessary conditions for forming a system with self-organization capability. A self-generation system is a kind of system with self-organization capability. The self-generation of machining precision for solid balls with super precision is emphatically explained. From the viewpoint of self-organization, there are three types of systems including system 1 with the self-regulation capability, 2 with the self-determination capability of goals, and 3 with the self-organization capability. Although they are all closed loop control systems, they have different constructions and functions. Necessary conditions for achieving self-generation of machining precision are given. Establishment of the system for machining solid balls with super precision is discussed. Self-generation of machining precision for solid balls with super precision on the basis of the capability of self-removal of errors is presented. Self-generation includes the ability of self-removal of errors for solid balls, convergence of self-removal of errors, self-generation of precision, and self-generating system for the given.

Abstract:

Based on an analysis of mechanism combination methods of a current mechanism system kinematic scheme, input/output kinematic behavior and their constraint relations were proposed to represent the kinematic behavior knowledge of a mechanism system. Furthermore, a tree structure of a kinematic behavior decomposition process for a mechanism system was provided. Considering multiple outputs for a mechanism system, the matching algorithm and the attributes propagation method of kinematic behavior were used to generate a mechanism combination scheme. Its intermediate design solution and the constraint relations between input and output are generated to fill the common blackboard. Moreover, using information in the blackboard as input motion, the behavior attributes of other process actions are transmitted to attribute items of the blackboard, which finally enables a computer-aided automatic design process of a mechanism system kinematic scheme. To avoid the problem of schemes combination explosion caused by unbounded depth in the search process, bounded depth-first search was used to control the number of expanded hierarchies for a design tree. Finally, an example was given to show its feasibility and solution efficiency.

Abstract: This study aimed to investigate the prevalence, clinical characteristics, and prognostic impact of 1p32.3 deletion in patients with newly diagnosed multiple myeloma (MM). A retrospective analysis was conducted on 411 patients with newly diagnosed MM; among which, 270 received bortezomib-based therapies, and 141 received thalidomide-based therapies. Fluorescence hybridization (FISH) was performed to detect six cytogenetic abnormalities, namely, del(1p32.3), gain(1q21), del(17p13), del(13q14), t(4;14), and t(11;14). Results showed that 8.3% of patients with MM were detected with del(1p32.3) and had significantly more bone marrow plasma cells ( = 0.025), higher 2-microglobulin levels ( = 0.036), and higher lactate dehydrogenase levels ( = 0.042) than those without del(1p32.3). Univariate analysis showed that patients with del(1p32.3) under thalidomide-based therapies (median PFS 11.6 vs. 31.2 months, = 0.002; median OS 16.8 vs. 45.9 months, <0.001) were strongly associated with short progression-free survival (PFS) ( = 0.002) and overall survival (OS) ( <0.001). Multivariate analysis revealed that del(1p32.3) remained a powerful independent factor with worse PFS ( = 0.006) and OS ( = 0.016) for patients under thalidomide-based treatments. Patients with del(1p32.3) under bortezomib-based treatments tended to have short PFS and OS. In conclusion, del(1p32.3) is associated with short PFS and OS in patients with MM who received thalidomide- or bortezomib-based treatments.
